International HapMap Project
The
goal of the International HapMap Project is to develop a haplotype map
of the human genome, the HapMap, which will describe the common patterns
of human DNA sequence variation. The HapMap is expected to be a key resource
for researchers to use to find genes affecting health, disease, and responses
to drugs and environmental factors. The information produced by the Project
will be made freely available.
The samples prepared for the project come from Asian people, European
people and African people. Among the samples, European samples come from
CEPH, China and Japan provided one half of Asian sample respectively while
African sample come from Nigeria. The sample collection stands to all
regulations of ethnics.
Introduction of the platform
Genotyping platform launched in Feb.2003 and equipped with FP of PerkinElmer
and mass spectrometry of SEQUENOM. Genotyping platform is working on establishing
Genotyping map of chromosome 3 and 21 of HapMap project.
Genotyping platform is young and vigorous and is composed of production
line, QC group and R&D group.
Production line
Production line equipped with two advanced examining technological platforms,
automatic working system and automatic bio-reaction system.The current
daily throughput of PerkinElmer system is 150 thousand reactions and output
of Sequenom system is 150 thousand Genotyping per day.
QC group
Newly founded in Dec.2003, QC group is indispensable in genotyping platform.
QC group is responsible for the quality control of production, reagents,
materials, instruments and platforms when working with PerkinElmer system
and Sequenom system. In addition, QC group takes charge of planning and
management of the project and statistic, analysis and collection of the
production data.
R&D group
Founded in Dec.2003, R&D group optimizes experiments in newly established
technology route, frames working flow, extends the output to large-scale
and keeps tracking the international developing trend of Genotyping technology
route.
Examining technique of genotyping platform
Fluorescence polarization detecting technology
In the reaction system, reacting with ddNTP that bearing deferent
fluorescence labeling, extension primer consequently takes deferent fluorescence
labeling. At the same time, as a result of combining with large molecule,
the degree of polarization of ddNTP changed.

Flying time mass spectrometry
By using of mass spectrometry, gene type was estimated since extension
primer extends one to several bases due to deferent position of SNP.

Yang Huanming and Zeng Changqing attended preparing meeting for
international HapMap project hold in NIH, US.
